The easiest way is to remove the left side cover and slide the spool out. Thread the line through the front of the reel. Tie the line to the spool. Insert the spool back into the reel. Start reeling. Be sure to keep the line tight. If your using braid, you will have to use a mono backer. There should be a diagram in the line box as to how to tie the two together. After a few times you can do it with your eyes shut. Hope this helps.

I would not recommend removing the spool, if you a spool with the holes in it i usually just put the line through the one of the holes and carefully start spooling the line after you get a couple of turns start pulling the line tight until it's completely tight and just continue spooling while keeping the line tight, I know some of you might say what if he gets spooled, I have never been spooled and you should never get spooled by a bass no matter how big it is.  I find this the easiest way to spool a baitcaster. Hope this helps.

 I have my reel on the rod and put the line thru the guide on the reel and use about 1/2 the tape that comes holding the line and tape it to the spool. Hold tension on the line about 1/2 way to first guide and wind with other hand. If it ever comes out uneven on the spool, it always corrects itself after the first cast or so...

I'm with CFFF, you don't need to remove the spool.  And if you have problems routing the line around the spool to tie an arbor knot, just use a dental pick or something to reach in and grab the line.  I used to use a dental pick, but now I use my slip ring pliers to grab the line.  Just a technique..... :)
