Okay, I gave a small spray bottle of this, how much should I spray on line that is wound on a spool already?

3-4 spritz around the entire spool is enough.  Let it dry.

This stuff makes a difference.

I usually spray it on my fluro before I fish it. Makes it more manageable.

Works well on mono too.

I give my mono and YZH this the night before a trip - don't use FC at all. Really makes a difference in both.

How long does it take to dry?

I use KVD line conditioner on the top 1/3rd of my line which is usually over mono backing . I spray a small cloth first then spool line on through my fingers rotating the cloth for a good application of the stuff . When done I spritz the spool with a few squirts of the stuff as well then let it sit over night ... Works well applied this way - don't over look braid either as it helps braid mainline last longer I believe .

Spray the reels the night before, and your good. While fishing, I've sprayed KVD 30 mins before using a reel with new line, copoly, and it worked just fine.

As long as water?  I suppose it would depend on the humidity, ventilation, and some other things.  At least an hour.  It's easiest for me to spray all my reels AFTER a trip.  That way they're ready for the next one.

I've made it part of my 'night before' checklist.

If your reels are already spooled though how does it get to the inner layers?

It's about as thin as water - so it seeps down through capillary action around the surface of the line. Just like pouring water over the spool, the lower layers will still get wet.

If you spray it on your teeth before bed, you'll have whiter teeth by morning.*

Maybe unrelated, Megastrike is pretty good for styling your hair.  A little drop of Gulp! juice on the wrists and behind the ears drives the ladies crazy, too.

L&L is one of the few things I completely believe in despite having no idea what the mechanism of action is.  No idea how it works and I honestly don't think I even want to know.

I do agree that you have to shake it up.  I buy in bulk and refill the little spray bottle.  It lasts forever, so this is like a once a year thing.  One year I neglected to shake my bulk bottle and filled the sprayer off the top of it.  I had a bad sticky time for a few days until I figured it out and got it poured back in and mixed properly.

The stuff just works.  I spray it on after I spool fresh line.  It works best on flouro and I apply it before every trip.

I want to buy a gallon, but they don't give the stuff away.

Do you have to use it every time you fish?

Necessary? No - every 3rd to 4th time probably will work.

Recommended? Yes - it's only a few spritz per reel, and even with 4 rigs last year that I used it on...barely dropped the level in the 16oz bottle I have. This year it's more - 7 of 10 rigs it'll be used on.

I walk into the yard and make a cast for this very reason...Just divide up a handful of sprays as new line stacks up when reeling in. I also  sometimes crank the line through a folded smooth sunglass cloth thats damp w/the stuff. You could probably put a little mark on the spool if you wanted to make sure your spraying opposing sides with ea spritz, but I never get that picky.
