If your using quality gear then IMO there isn't a noticeable difference. If you want a two piece rod because it is more convenient for you then I would get one!

IMO there is a difference but it is minor. 2 piece tend to be a bit softer in general. Get the 2 piece if preferred.

You will not be able to tell the difference if both rod are about the same price and quality.

There has been a major improvement in two piece casting rods over the last decade. Prior to that, I wouldn't recommend one.  Look for a quality brand that offers both. The so called dead spot where the two pieces join on a casting rod should be somewhere between the first and second guide (from the handle). In that position, it won't effect sensitivity or the loss of any power enough for most anglers to notice.

These aren't your grandfather's 2 piece rods anymore.  there is much technology in the ferrules that make them perform as well as 1 piece rods.

I found just the opposite when comparing the St. Croix 3s76mlxf and the 3s76mlxf-2.  I built the 1 piece for myself and the 2 piece for a friend.  The 2 piece had a touch more power, which I really liked.  I'm gonna sell the 1 piece and build myself the 2 piece when I find the time.

As long as you stay with the same level of quality multi piece rods are every bit as good as their one piece counterparts. Scott is right about two piece having a touch more power. My guess is from the double wall where the ferrules are.
